It's kind of an "in joke" now on twitter. Trending topics don't get picked up by hashtags anymore, or so I'm told.
The "@" is a username, so if I were to mention you (and your username was "LordKangana") by putting @LordKangana, you would see the message in your "mentions" column, rather than *just* in your "feed".

"at" sign is your user name.
My username = @JebusF
If you wanted to message me, in your tweet you would write "@JebusF i am messaging you"
As in "my message is aimed @JebusF"
The hashtags are to group together messages related to that tag. IE, if you say "@JebusF I am messaging you about my #penis" then you could click the hashtag (#penis) and see messages that OTHER users have written which also includes "#penis".
Another example "Shit, shocking injury to Stuart Holden #bwfc" means that other fans of #bwfc can click it, see all message including "#bwfc"
